摘要:Objective To establish a method of reducing rabbit bone marrow mesenchymalstem cells (MSCs) to express chondrocytic phenotype. Methods MSCs were seperated from bonemarrow ertracted from the iliac of New Zealand rabbit. TGF-β1, IGF- I and Vitamin C were appliedinto culture medium to indntee proliferation and transformation of MSCs. Colony forming efficiency( CFE) was measured to reflect cell proliferation rate; procollagen al ( Ⅱ )mRNA in cells was detectedby RT- PCR ; ultrastructure of cells was observed under scanning electronic microscope ( SEM ) ; alklinephosphatase(ALP ) in cullure medium wts also detected. Results Under the influence of TGF-β1,IGF-1 and Vitamine C, CFE of MSCs rose from control level of 3/106 to 21/106, expression ofarticular cartilage specific procollagen al ( Ⅱ )mRNA by cells was promoted; At the same time, floccu-lent matrixes synthesized were observed between connecting polygonal MSCs. ALP in culture medium de-clined to control level with culture time. Conclusion Expression of chondrocytic phenotype byMSCs could be induced by the synergistic action of TGF-β1, IGF- I and Vitamine C. Growth factor-intduced MSCs could be a good cell source for tissue-engineered cartilage.
