摘要:The rat calvarial bone defect model is widely applied in bone tissue engineering, especially in the animal model for bone grafting materials and relative cytokines. There are many disputes on the diameter size of the critical size defects ( CSD ), unilateral or bilateral defects, whether to shield the dura mater and pericranium during the process of establishing the rat calvarial bone defect model. Based on our literature review, there are many different disputes about the species, sex, and age of rats. We've confirmed that male adult ( 10 - 12 weeks, 300 - 350g , Sprague Dawley / SD ) rats are preferred in the model establishment. When the CSD is 5 mm, the researcher can make a self-comparison and avoid the damage of the sagittal suture, thus reduce the number of rats with the bilateral defects at the same time. When the CSD is 8 mm, a larger range of defects may be established, so as to make the experimental results more convincing. The dura mater and pericranium can promote the repair of calvarial bone defects as intramembranous bone growth sites. Therefore, we may consider shielding the dura mater and pericranium during the process of evaluating the bone repair materials and relative cytokines to avoid the interference on the results.
