摘要:OBJECTIVE:To investigate the structure,location,function and regulation of Zonula occluden-1(ZO-1)aswell as the correlation of ZO-1 with diseases for helpling diagnosis,treatment and determining prognosis of correlative diseases.DATA SOURCES:Using the terms"Zonula Occluden"we searched the Medline database to identify ZO-1 related articles published between May 1963 and December 2006 in English.STUDY SELECTION:The articles were selected firstly,and those involved the structure,location,function and regulation of ZO-1 as well as the correlation of ZO-1 with diseases were chosen.The full texts of the left articles were searched.Inclusive criteria:Studies associated with the structure,location,function and regulation ofZO-1 as well as those associated with the correlation of ZO-1 with diseases.Exclusive criteria:Studies withrepetitive or uncorrelated contents. DATA EXTRACTION:Among 1 462 collected articles of ZO-1,1 424 were excluded.of the 38 articles.whichmet inclusive cdteria,11,2,10,11 and 4 were associated with the structure,location,function and regulation ofZO-1 as well as the correlation of ZO-1 with diseases.respectively.DATA SYNTHESIS: Z0-1 is a member of membrane associated guarylate kinasellike proteins (MAGUKS).can not form homooligomers and exists as ZO-1/ZO-2 and ZO-1/ZO-3 heterodimers.It is expressed in endothelial cells and epithelial cells and also in other types(except for tight iunction)of cellS.It exists not only intight junction but also in adherens junction."Messenger"which contacts with various iunctional complexes,interacts with tight junction,adherens junction and gap junction to maintain the structure and function of iunctional complex.The structure and function of ZO-1 are regulated by many factors,such as Ca2+,phosphorylation.protein kinase C,G protein and so on.ZO-1 plays an important role in the treatment of ischemia/hypoxia.inflammation and tumors. CONCLUSION:ZO-1,which is only used as a tight junction protein initially,has been found to be a"messenger"which can contact various junctional complexes,interacts with tight junction,adherens junction and gap junction and is related with the pathogenesis of many diseases.
