Expression of pulmonary activation regulated chemokine in acute paraquat poisoning
Abstract:Objective This study was conducted in order to elucidate the expression of pulmonary activation regulated chemokine ( PARC) in lung injury by acute poisoning of paraquat and to determine whether PARC can be used as an effective index of pulmonary fibrosis evaluation of paraquat poisoning. Methods A total of 90 healthy adult rats were randomly divided into paraquat exposure experimental group and saline control group .The experimental rats by intraperitoneal injection of paraquat solution (20 mg/kg).Lung tissue, blood and bronchoalveolar lavage fluid (BALF) were sampled at 6 time points:1, 3, 7, 10, 14 and 21 d after poisoning.Pathological changes of lung tissue were observed by HE staining, classified cells were counted in BALF and ELISA method was used to detect the concentration of PARC in blood and BALF .Results The macrophages in BALF from 3 d were gradually increased in the experimental group , compared with the corresponding time point of saline control group (P <0.05).The concentration of PARC of experimental group in BALF and blood from 3 d was significantly higher than that in the control group , and the difference was statistically significant ( P<0.05 );and with the time prolonging , the content of PARC in bloodin creased gradually .Conclusion In the process of paraquat poisoning , the concentration of PARC can effectively assess the progress degree of pulmonary fibrosis .
