Study on the Synthesis and Practical Application of Chitosan-silver Nanoparticles Targeting Pseudomonas Aeruginosa Biofilms

摘要:Objective:To develop chitosan-silver nanoparticles targeting Pseudo-monas aeruginosa biofilms and verify their antibacterial performance through animal experiments.Methods:Chitosan,silver nitrate,glacial acetic acid,and other chemical reagents were used to synthesize chitosan-silver nanoparticles.The characterization,minimum inhibitory concentration,and biofilm inhibition rate of the chitosan-silver nanoparticles were tested.A total of 40 SD rats were randomly divided into four groups.After routine adaptive feeding,the control group received intraperitoneal injection of normal saline;the model group received intraperitoneal injection of Pseudomonas aeruginosa suspension;the positive group received intraperitoneal injection of Pseudomonas aeruginosa suspension mixed with ampicillin at a volume ratio of 1∶1;the observation group received intraperitoneal injection of Pseudomonas aeruginosa suspension mixed with chitosan-silver nanoparticles(at minimum inhibitory concentration)at a volume ratio of 1∶1.Bacterial load,inflammatory factors,and liver and kidney function indicators in tissues were observed and compared among the four groups on the 3rd day after treatment.Results:When the concentration of chitosan-silver nanoparticles reached 8 μg/mL or above,the OD value of the experimental wells was close to that of the control wells,indicating that 8 μg/mL was the minimum inhibitory concentration of the chitosan-silver nanoparticles;at concentrations of 8 μg/mL or above,the biofilm inhibition rate was greater than 80%.The bacterial load in the observation group was significantly lower than that in the model and positive groups(P<0.05).The expression levels of interleukin-6,interferon-γ,and tumor necrosis factor-α in the observation group were significantly lower than those in the model and positive groups(P<0.05).There were no statistically significant differences in alanine aminotransferase,aspartate aminotransferase,blood urea nitrogen,and creatinine levels among the four groups(P>0.05).Conclusion:The chitosan-silver nanoparticles targeting Pseudomonas aeruginosa biofilms constructed in this study exhibit good antibacterial effects against Pseudomonas aeruginosa and have good safety.

The Regulatory Effect of miR-146a Overexpression on Corneal Inflammatory Response in a Mouse Model of Dry Eye Disease and Its Relationship with the IRAK1/TRAF6/NF-κB Signaling Pathway

摘要:Objective:To investigate the regulatory effect of miR-146a overexpression on corneal inflammatory response in a mouse model of dry eye,and to analyze its relationship with the IRAK1/TRAF6/NF-кB signaling pathway.Methods:A total of 50 SPF-grade BALB/c mice were randomly divided into five groups,with 10 mice in each group.Except for the control group,the other four groups were treated with 0.2%benzalkonium chloride(BAC)solution in both eyes to construct a dry eye model.After successful modeling,the control group and model group received NC agomir;the miR antagonist group received miR-146a antagomir;the miR agonist group received miR-146a agomir;and the pathway agonist group received miR-146a agomir+NF-κB activator 2.After four weeks of treatment,the expressions levels of miR-146a,inflammatory factors,and IRAK1/TRAF6/NF-κB signaling pathway proteins were observed and compared among the five groups.Results:After four weeks of treatment,there was a statistically significant difference in the relative expression of miR-146a in the five groups(F=61.058,P<0.001),which was significantly higher in the miR agonist group than in the other four groups.After 4 weeks of treatment,there were statistically significant differences in the expression levels of IL-1β,IL-6,IL-8 and TNF-α in the five groups(F=84.757,103.658,55.477,46.762;P<0.001).After four weeks of treatment,there were statistically significant differences in the protein expression levels of IRAK1,TRAF6,NF-κB and IκB α in the five groups(F=62.975,77.173,67.108,29.381;P<0.001),except for the control group,the expression levels of IRAK1,TRAF6 and NF-κB proteins in the miR agonist group were significantly lower than those in the other three groups,and the expression levels of IκBα protein were significantly higher than those in the other three groups.Conclusion:Overexpression of miR-146a can negatively regulate corneal inflammatory response in dry eye mice through the IRAK1/TRAF6/NF-κB signaling pathway,which can provide new insights for the clinical treatment of dry eye disease.

Experimental Study of a Novel Stone-dissolution Controlled-release System for Urinary Tract Stones Combined with Urinary Tract Infection

摘要:Objective:This study aims to develop a drug controlled-release sys-tem based on nano-hydrogel technology for stone dissolution and evaluate its dual effi-cacy in synchronously dissolving calcium oxalate stones and inhibiting uropathogenic Escherichia coli.Methods:A pH-responsive chitosan(CS)-sodium alginate(SA)-hy-drogel microsphere loaded with potassium sodium hydrogen citrate and fosfomycin(CS/SA@PSHC@CS@F)was constructed using ionic cross-linking.In vitro stone dis-solution experiments were conducted to determine drug release kinetics and stone mass reduction rate under different pH environments,and the antibacterial activity a-gainst E.coli ATCC 25922 was evaluated using the plate counting method.A rat mod-el of kidney stone combined with infection was established.Stone volume changes were quantified via micro-CT,urinary interleukin-6(IL-6)and tumor necrosis factor α(TNF-α)levels were measured by ELISA,and renal tissue inflammation was scored via histopathological evaluation.Results:The 24-hour cumulative release rate of CS/SA@PSHC@CS@F at pH 5.8 reached 92.3%±4.1%,significantly higher than at pH 7.4(31.2%±3.7%,P<0.01).At 72 h,the mass reduction rate of calcium oxalate stones in CS/SA@PSHC@CS@F was 68.9%±5.2%,significantly higher than that of the PSHC+F mixture(P<0.01).Antibacterial experiments confirmed that the inhibition rate of biofilm colonies by CS/SA@PSHC@CS@F could reach 82.4%±6.7%,compared to 58.2%±5.3%for F,with a statistically significant difference(P<0.01).Animal experiments showed that the stone volume reduction rate in the CS/SA@PSHC@CS@F group was 62.3%±8.1%,higher than that in the PSHC+F group,and the decrease in urinary IL-6 level after administration was more significant,while there was no significant differ-ence in TNF-α level compared with the other two groups(P>0.05).The renal tissue inflammation score decreased to 2(1,2)points,showing a significant reduction com-pared with the other two groups(P<0.05).Conclusion:The novel controlled-release system designed in this study achieves targeted drug release in the stone infection mi-croenvironment through the ion exchange mechanism,simultaneously enhancing stone dissolution efficiency and antibacterial effect,thereby providing a new drug delivery strategy for the treatment of urinary stones combined with infection.

Study on the Impact of Single-port Laparoscopic Surgery Based on Minimally Invasive Instrument Engineering Optimization on Biomarkers of Ovarian Reserve Function

摘要:Objective:To investigate the quantitative influence of engineering parameters(diameter,bending angle)of minimally invasive surgical instruments and single-port laparoscopic surgery on biomarkers of ovarian reserve function,and es-tablish a multi-parameter coupled mathematical prediction model,providing a theo-retical basis for precision instrument design and operation optimization.Methods:A total of 45 reproductive-age patients undergoing single-port laparoscopic ovarian cystectomy were selected.Through orthogonal experimental design,the instrument di-ameter(3/5/8 mm)and bending angle(30°/60°/90°)were divided into 9 groups,with 5 cases in each group.Changes in serum AMH(anti-müllerian hormone),FSH(folli-cle-stimulating hormone),INHB(inhibin B)and ultrasonic AFC(antral follicle count)indexes were measured before surgery and on the 2nd to 3rd day of the first men-strual period after surgery.Enhanced CT images and laparoscopic images of ovarian force distribution were obtained.Finite element analysis(FEA)of the single-port in-strument-ovarian tissue model was performed using ABAQUS software to calculate stress distribution in the ovarian cortex.Oscillatory shear experiments were used to determine the characteristics of viscoelastic property changes in the linear viscoelastic range of ovarian tissue.Results:The rate of decrease in AMH was lower in the 3 mm diameter-30°angle group(P<0.05);the peak Von Mises stress in the D3-θ30 group was lower than that in the other groups(P<0.05);the optimal parameter combination was a diameter of 3.13 mm and an angle of 21.72°,with the model calculating an AMH attenuation of 20%.Ovarian tissue changed with the frequency of the oscillatory shear test.In the low-frequency region(<1 Hz):G'>G''(elasticity-dominated);in the high-frequency region(>5 Hz):G''>G'(viscosity-dominated);the crossover point was at 5 Hz,where G'=G''=5 kPa.Before ovarian dissection:G'was relatively high and G''was relatively low,with elasticity dominating.During dissection:G'decreased and G''increased,with instrument stress causing damage.After dissection:G'further de-creased and G''continued to rise,resulting in irreversible damage to the ovarian stro-ma.Conclusion:The engineering parameters of minimally invasive instruments signifi-cantly affect the ovarian reserve function index AMH and the peak Von Mises stress of the ovary through mechanical stress transmission.Using an instrument with a diameter of 3.13 mm and a bending angle of 21.72° can more effectively reduce the impact of single-port laparoscopic surgery on AMH decline.

Screening Differential Protein Pathways in Insulin Resistance Complicated with PCOS Using iTRAQ Combined with LC-MS/MS Technology

摘要:Objective:To screen potential serum differential proteins in insulin re-sistance(IR)complicated with polycystic ovary syndrome(PCOS)using isobaric tags for relative and absolute quantitation(iTRAQ)combined with ultra-high performance liquid chromatography-tandem mass spectrometry(LC-MS/MS).Methods:A total of 20 pa-tients diagnosed with PCOS in our hospital were selected,including 10 cases of simple PCOS and 10 cases of PCOS+IR.iTRAQ combined with LC-MS/MS was used for pro-teomic analysis to identify serum differential proteins.Bioinformatics analyses,including gene ontology(GO),Kyoto encyclopedia of genes and genomes(KEGG),and pro-tein-protein interaction(PPI)analysis,were conducted to understand the biological pro-cesses,cellular components,and molecular functions of the differentially expressed pro-teins detected by the two methods.Results:A total of 454,675 secondary spectra were detected by iTRAQ,with 14,376 matched spectra.Combined with LC-MS/MS,74,386 peptides,47,542 unique peptide sequences,and 54,675 proteins were identified.A total of 249 differentially expressed proteins with a fold change≥1.30 or≤0.83 were found,among which 9 had a P-value<0.05.There were 5 up-regulated and 4 down-regulated proteins,namely RPAP3,ALDH6A1,COX20,RASSF3,ALPK2,NANOS1,FAM210A,CHGA,and CGA.These differential proteins were imported into the STRING database for PPI network analysis,which revealed 21 nodes and 69 edges with a PPI enrichment P-value<0.001.KEGG enrichment results included hsa04913(Ovarian steroidogene-sis),hsa04024(cAMP signaling pathway),and hsa04080(Neuroactive ligand-receptor interaction).Conclusion:The combination of iTRAQ and LC-MS/MS technologies i-dentified nine differential proteins including CHGA and CGA,which are mainly en-riched in pathways related to ovarian steroidogenesis,cAMP signaling,and neuroactive ligand-receptor interaction.This combination provides a powerful platform for exploring the pathogenesis of IR complicated with PCOS and discovering new biomarkers.

Construction of pH-Responsive Paclitaxel-exosome Composite Nanocarriers and Their Inhibitory Effect on the Proliferation of Endometrial Cells

摘要:Objective:To construct a pH-responsive paclitaxel(PTX)-exosome composite nanocarrier and investigate its inhibitory effect on the proliferation of endometrial cancer cells(HEC-1A).Methods:PTX was loaded into exosomes derived from adipose mesenchymal stem cells using the thin-film hydration method,and modified with polyethylene glycol-polylactic-co-glycolic acid(PEG-PLGA)to form nanocarriers(PTX-Exo-NPs).The particle size and morphology were detected by nanoparticle size and Zeta potential analyzer;drug encapsulation efficiency and drug loading capacity were determined by high-performance liquid chromatography;drug release behavior was evaluated in vitro under simulated acidic(pH 5.5)and physiological(pH 7.4)conditions;MTT assay and flow cytometry were used to detect the effects of the carrier on the proliferation,apoptosis,and cell cycle distribution of HEC-1A cells.Results:PTX-Exo-NPs exhibited a uniform spherical shape with a particle size of(128.5±5.2)nm,PTX encapsulation efficiency of 92.3%±2.1%,and drug loading capacity of 15.6%±0.8%.Drug release rate in the acidic environment(85.3%±2.1%within 72 h)was significantly higher than that in the physiological environment(48.0%±1.7%).In vitro experiments demonstrated that the proliferation inhibition rate of PTX-Exo-NPs on HEC-1A cells was higher than that of free PTX,with a lower IC50(0.64 μM vs 4.70 μM),and could induce cell apoptosis(apoptosis rate:28.7%±2.1%vs 14.2%±1.5%)and promote cell cycle arrest(G2/M rate:45.3%±3.2%).Conclusion:PTX-Exo-NPs exhibit pH-responsive characteristics,which can target drug release through the acidic microenvironment,enhance the proliferation inhibition and pro-apoptotic effect on endometrial cancer cells,thus serving as a potential strategy for targeted therapy of endometrial tumors.

Neural Network Based on GA-BP Algorithm and its Application in the Protein Secondary Structure Prediction

摘要:The advantages and disadvantages of genetic algorithm and BP algorithm are introduced. A neural network based on GA-BP algorithm is proposed and applied in the prediction of protein secondary structure, which combines the advantages of BP and GA. The prediction and training on the neural network are made respectively based on 4 structure classifications of protein so as to get higher rate of predication-the highest prediction rate 75.65%, the average prediction rate 65.04%.

Analysis of Direct Volume Rendering in VTK

摘要:This paper introduces the techniques of direct volume rendering in VTK and discusses ray-casting-the most popular technique of direct volume rendering. The visualizations for medical volumetric data by three functions of raycasting are offered. Visualization effects prove that VTK is easy to get satisfied results with good quality in short time.

A Community Health Service Architecture Based on the Internet of Things on Health-Care

摘要:In order to assure a real-time medical care for the elders who live alone or have a chronic disease and also to improve the responsiveness and quality of the community hospital, a community health service architecture was designed according to the Internet of Things on Health-Care. The users' body physiological and sports information can be collected intelligently by the health sensors at home and then the health information can be transmitted to the community health-care center through networks after information processing. The community doctors can provide a medical service on kinds of video, voice or email according to the users' process notes, test data and symptoms. Of course, doctors also can communicate and share their opinions with other specialist who is online. The key technology of this system is Internet of Things on Health-Care, which system can get users' health information actively and then make out intelligent decisions promptly and finally provide a more personal health service.

The Establishment and Development of Finite Element Model of Human Cervical Vertebra and Its Application Example

摘要:.The incidence rate of cervical spondylosis is high,and due to the complicacy of cervical vertebra structure, irregularity of shapes and non-uniformity of components, sometimes it's difficult to achieve planned objectives by experiments in vitro through stress and strain analysis. Besides, the biomechanical factors are of vital significance in the cause of spinal disorders. In this paper the author makes a summary of the present modeling of human cervical vertebra and describes the major methods of establishing the finite element model of human cervical vertebra through several self-constructed models. With the advance of computer technology, the finite element methods have been rapidly developed in cervical vertebra biomechanical researches and have became a major approach for biomechanical researches to simulate more and more clinical conditions.

A Study on the Temperature Correlation of B-mode Ultrasonic Image Gray for Noninvasive Temperature Monitoring in Hyperthermia

摘要:This paper deals with the temperature correlation of gray scale of Bmode ultrasound image from heated tissue. In this study, many in-vitro fresh pig livers are heated in a temperature range from 28℃ to 45℃, from which a series of B-mode ultrasonic images of livers were obtained. The gray-value is evaluated from the ultrasound images respectively. A correlation of the mean gray value of the selected regions (12×12 pixels) in B-mode ultrasonic images of liver and its temperature was pointed out. And the experiment results agreed the evaluation well. And it is possible to monitor the tissue temperature changing in hyperthermia using this correlation.

Expression and Subcellular Localization of Recombinant SDF-1 and Its Mutant Intrakine in Transfected COS-7 Cells

摘要:Objective:This paper is to explore a method of transferring human SDF-1 and its mutant SDF-1/54 intrakine gene into COS-7 cells for determining their expression and subcelluar localization of the fusion protein. This could offer feasibility for inhibiting the metastasis of malignant tumors by phonotypic knockout for blocking functional expression of receptor on the cell-surface. Methods:Amplify the target gene with PCR from the constructed plasimid SDF-WT-Gly×4-Dec/PET-30a(+)with a C-terminal retention signal fragment KDEL. After the pcDNA3.1 /SDF-1/KDEL, pcDNA3.1 /SDF-1/54/KDEL, pEGFP/SDF-1/KDEL and pEGFP/SDF-1/54/KDEL eukaryotic expression vectors were constructed and the DNA sequence was accurate, they were transferred into COS-7 cells with liposome. The exogenous expressions were observed, fusion protein SDF-1/His and SDF-1/54/His were confirmed by Western blot, and the SDF-1/EGFP and SDF-1/54/EGFP were determined by Laser Scanning Confocal Microscopy. Results:Four expression vectors were constructed successfully, the fusion protein SDF-1/KDEL/His and SDF-1/54 KDEL/His expressed in COS-7 cells. Subcelluar localization analysis showed that SDF-1/KDEL/EGFP and SDF-1/54/KDEL/EGFP were located mainly in endoplasmic reticulum. Conclusion: Four expression vectors pcDNA3. 1/ SDF -1/KDEL, pcDNA3.1/SDF - 1/54/KDEL, pEGFP/SDF - 1/KDEL and pEGFP/SDF-1/54/KDEL were constructed successfully, which could express in eukaryotic cell and locate mainly in the endoplasmic reticulum.

Wavelet Variance Analysis of EEG Based on Window Function

摘要: A new wavelet variance analysis method based on window function is proposed to investigate the dynamical features of electroencephalogram (EEG).The ex-prienmental results show that the wavelet energy of epileptic EEGs are more discrete than normal EEGs, and the variation of wavelet variance is different between epileptic and normal EEGs with the increase of time-window width. Furthermore, it is found that the wavelet subband entropy (WSE) of the epileptic EEGs are lower than the normal EEGs.

Removal of White Noise from ECG Signal Based on Morphological Component Analysis

摘要:To effectively suppress white noise and preserve more useful components of electrocardiogram (ECG) signal, a novel de-noising method based on morphological component analysis (MCA) is proposed. MCA is a method which allows us to separate features contained in an original signal when these features present different morphological aspects. According to the features of ECG, we used the UWT dictionary to sparsely represent mutated component, and used the DCT dictionary to sparsely represent smooth component. The experimental results of the samples choosing from MIT-BIH databases show that the MCA-based method is effective for white noise removal.

Four-Dimensional Computerized Tomography (4D-CT) Reconstruction Based on the Similarity Measure of Spatial Adjacent Images

摘要:Objective:To investigate the feasibility of a 4D-CT reconstruction method based on the similarity principle of spatial adjacent images and mutual information measure. Methods:A motor driven sinusoidal motion platform made in house was used to create one-dimensional periodical motion that was along the longitudinal axis of the CT couch. The amplitude of sinusoidal motion was set to an amplitude of ±1 cm. The period of the motion was adjustable and set to 3.5 s. Phantom objects of two eggs were placed in a Styrofoam block, which in turn were placed on the motion platform. These objects were used to simulate volumes of interest undergoing ideal periodic motion. CT data of static phantom were acquired using a multi-slice general electric (GE) LightSpeed 16-slice CT scanner in an axial mode. And the CT data of periodical motion phantom were acquired in an axial and cine-mode scan. A software program was developed by using VC++ and VTK software tools to resort the CT data and reconstruct the 4D-CT. Then all of the CT data with same phase were sorted by the program into the same series based on the similarity principle of spatial adjacent images and mutual information measure among them, and 3D reconstruction of different phase CT data were completed by using the software. Results:All of the CT data were sorted accurately into different series based on the similarity principle of spatial adjacent images and mutual information measures among them. Compared with the unsorted CT data, the motion artifacts in the 3D reconstruction of sorted CT data were reduced significantly, and all of the sorted CT series result in a 4D-CT that reflected the characteristic of the periodical motion phantom. Conclusion:Time-resolved 4D-CT reconstruction can be implemented with any general multi-slice CT scanners based on the similarity principle of spatial adjacent images and mutual information measure.The process of the 4D-CT data acquisition and reconstruction were not restricted to the hardware or software of the CT scanner and has the feasibility ,which extensive applicability.
