晚期软组织肉瘤的靶向治疗

摘要：Soft tissue sarcoma(STS)is a rare but highly heterogeneous group of mesenchymal-derived solid tumors comprising various pathological subtypes,with poor prognosis in advanced stages.The current first-line treatment for advanced STS is anthracycline-based chemotherapy,which has limited efficacy,significant toxic side effects,and poor response in certain STS subtypes.In addition to chemotherapy regimens selected based on pathological subtypes for second-line treatment,both Chinese and international clinical guidelines also recommend the use of anti-angiogenic targeted therapies.With the deepening understanding of molecular mechanisms of STS,other types of low-toxicity and high-efficiency targeted drugs have gradually entered the field of advanced STS treatment.Based on the basic characteristics of tumor biology,this review systematically examines targeted therapies currently entering clinical research on advanced STS from two aspects:targeting tumor cell signaling pathways and targeting the tumor microenvironment.Specifically,this article introduces the action mechanisms,clinical trial results,approved indications and clinical availability.This review aims to explore the current status and research progress of targeted therapy in advanced STS,and provide reference for more scientific and standardized use of targeted drugs to improve the therapeutic efficacy and prognosis of patients with advanced STS.
